Investigation of Anti-aging Effect and Determination of Chemical Structure of Pine Needle (Pinus denstflora) through Animal Experiment ¥². Effects of Butanol Fraction on Oxygen Radicals and Their Scavenger Enzymes in Brain of SD Rats

Abstract
Oxigen radicals and their scavenger enzymes in brain membranes of SD rats were investigated on a butanol fraction of pine (Pinus dens flora Sieb et Zucc.) needle. SD rats were fed experimental diets prepared with BuOH of 25, 50 and 100 mg/kg BW/day for 6 weeks. Cholesterol accumulations were significantly inhibited (13.6% and 16.8%, 19.7% and 30.4%, respectively) in brain mitochondria and microsomes of BuOH-50 and BuOH-100 groups compared with control group. In vitro, BOR formations were significantly inhibited (about 8-12% and 17-20%, respectively) in brain mitochondria and microsomes of BuOH-50 and BuOH-100 groups, but IOR formations were remarkably inhibited about 11-19% in motochondria of BuOH-50 and BuOH-100 groups compared with control group. Hydroxyl radical (OH) were significantly inhibited about 9-10% in mitochondria of BuOH-50 and BuOH-100, and 15-25% in microsomes of BuOH-50 and BuOH-100 groups compared with control group. Mn-SOD and GPx activities were significantly increased about 13.3% and 11.5% in brain membranes of BuOH-100 group compared with control group, but significant difference between Cu/Zn-SOD activities could not be obtained. These results suggest that butanol fraction of pine needle also may play an effective role in an attenuating oxygen radicals and activating scavenger enzymes, thus aging may be effectively modulated.
